In-situ studies with electronic monitoring of feedingbehaviour. Pymetrozine, a pyridine azomethine compound, representsa novel insecticide with a selective activity against homopteraninsects. It acts in a unique way: aphids are not knocked down oncontact but seem to die of starvation. This implies an effect ofPymetrozine on feeding behaviour. The aim of the present work is toelucidate how and at which step Pymetrozine interferes with thecomplex mechanisms underlying phloem feeding. The effect ofPymetrozine, applied in various ways, on different phases of styletpenetration and feeding activity of individual aphids is studiedusing the Electrical Penetration Graphue (EPG). Initialchoice experiments indicated that Pymetrozine does not have adeterrent or antifeedant action. Topical application (150 ng/mgfresh weight) inhibits stylet insertion into the plant. Wheninjected, less than 30 ng/mg is sufficient to produce the sameeffect. When Pymetrozine is systemically applied via plant sprayingor root uptake, aphids started feeding normally. After some time,however, they withdrew their stylets from the phloem and walkedaround with unaffected locomotion. At low doses aphids eventuallyrecovered and resumed feeding. High doses, however, irreversiblydisrupted feeding and prevented stylet reinsertion. Aphid motilityis not affected up to an estimated haemolymph concentration of 1 mMPymetrozine.
